Capabilities of the laboratory

The laboratory of metabolic studies at 57357 enables a series of metabolic, spectrophotometric, fluorescence, and EPR assays that measure selected mitochondrial respiratory function, metabolic pathways including oxidative phosphorylation, glycolysis, fatty acid oxidation, and amino-acid oxidation. It also allows EPR spin trapping assays to detect and quantify reactive oxygen species in fresh samples including isolated mitochondria and attached as well as suspended cells to assess membrane fluidity. The selected methods are carefully geared to enable translational as well as basic research in the field of cancer biology and other fields as well.

Main equipment/ Tech used in the lab

The following is a list of equipment/facilities that are available and running in our laboratory:

  • Oroboros O2k Station (Oroboros Inc., Austria) equipped with fluorescence modules that can assess H2O2, Ca2+, mitochondrial transmembrane potential, etc.

  • Seahorse XFe96 Analyzer (Agilent). Capable of performing metabolic analyses of upto 96 different conditions. Multiple parameter simultaneous measurements provide a systems-level view of cellular metabolic function in cultured cells and ex-vivo samples. The system is the spearhead in drug discovery efforts aiming at identifying drug candidates against cancer cells.

  • Cytation 5 Cell Imaging Multi-Mode Reader (Biotek, U.S.A.). The system combines fluorescence and high contrast brightfield imaging with conventional multi-mode detection without the expense and complexity commonly associated with other digital microscopy systems. It provides both quantitative phenotypic cellular information with well-based quantitative data.

  • Benchtop Magnettech Electron Paramagnetic Resonance (EPR) MS-5000 Spectrometer with gas and temperature control accessories. This system is the only technology that allows the detection, identification, and quantification of reactive oxygen species. It also allows the study of membrane biophysical characteristics among many other biological parameters.
